Introduction

Chemiluminescence immunoassay (CLIA) is a novel immunoassay technology that integrates highly sensitive chemiluminescence reaction and highly specific immune recognition to perform quantitative or qualitative analysis of antigens or antibodies. CLIA was first reported by Aiakawe in the mid-1970s. Now, it has been used as a mature and advanced technology for detecting trace amounts of active substance. CLIA consists of two parts: the immune response system and the chemiluminescence analysis system. In the immune response system, luminescent substance is directly labeled on antigens or antibodies (by using reactants to generate an excited-state intermediate), or acts on chemiluminescent substrates by means of enzymes. In the chemiluminescence analysis system, the chemiluminescent substance is catalyzed and oxidized to form an excited-state intermediate. When returning to a stable ground state, the excited-state intermediate emits photons. Given that the yield of photons is in direct proportion to the amount of the substance under test, the content of the substance in the sample can be calculated by establishing a standard curve.
Objective

CLIA integrates the sensitivity of chemiluminescence and the specificity of immune response. As a new immunoassay technology for ultramicro determination, it is characterized by high sensitivity, a wide range of linearity, stability and automation. CLIA applies to the determination and analysis of antigens, haptens, antibodies, hormones, enzymes, fatty acids, vitamins and drugs in the fields such as biomedicine, veterinary medicine and food safety testing.
Classification

CLIA can be divided into three categories based on the type of markers：
1、Direct chemiluminescence immunoassay: acridinium esters are used to directly label antibody (antigen) and react with the corresponding antigen (antibody) in the test sample to form a complex consisting of the solid-phase coated antibody, the antigen under test and the antibody labeled with acridinium esters. Next, it is only necessary to create an alkaline environment by adding add oxidant (H2O2) and NaOH. Acridinium esters are able to decompose and emit light in the absence of catalyst.
2、Chemiluminescence enzyme immunoassay (CLEIA): Antigen or antibody is labeled with the enzymes capable of catalyzing chemiluminescence reactions, such as horseradish peroxidase (HRP) or alkaline phosphatase (ALP). After the immune reaction with the corresponding antigen (antibody) in the test sample, they form a complex consisting of the solid-phase coated antibody, the antigen under test and the antibody labeled with enzymes. The substrate (luminescent agent) is added after washing, and catalyzed and decomposed by enzymes to emit photons. The photons are received by a special reading system as optical signal. The optical signal is converted to electric signal and amplified in a photomultiplier. The amplified electric signal is finally transmitted to the data processing system for calculating the concentration of the measured object.
3、Electro-chemiluminescence immunoassay (ECLIA): It uses RuCbpy (the electro-chemiluminescence agent) to label the antibody (antigen), and selects tripropylamine (TPA) as the electron donor. Specific chemiluminescence reaction occurs due to the electron transfer in an electric field. The reaction consists of two processes: electrochemistry and chemiluminescence.

Characteristics

A complete set of stable CLIA production process system has been established in our company. For optimization of core raw material selection, solid phase coated vectors, chemiluminescent markers, coating and labeling, reaction systems and quality control systems, etc., a strictly standardized process has been also set up to ensure stable performance of our products. At present, we succeed in developing a chemiluminescence enzyme immunoassay based antigen dectection kit on the existing process platform. The kit has the following remarkable features：
1、Sensitivity: 100 times greater than that of ELISA and fluorescence methods;
2、Specificity: Background noise can be removed;
3、A wide range: Luminescence intensity lie in 4~6 magnitudes, which is linearly correlated with material concentration to be determined;
4、Rapidness: Detection can be completed within 30-90 minutes;
5、High efficiency: Automation can be realized and high detection efficiency is achieved;
6、Stability: The period of validity reaches 12 months and above.
